Vanadate blocks cyclic AMP-induced Stimulation of sodium and water transport in amphibian epithelia

Abstract

INTEREST in the biological effects of vanadium has grown particularly since Cantley et al.1 identified vanadate as a potent inhibitor of (Na+ + K+) ATPase and Balfour et al.2 reported its striking natriuretic and diuretic properties in the rat. Such findings prompted us to investigate the effects of vanadate on amphibian epithelia, a model widely used for studying, in vitro, the membrane transport of sodium and water3,4. We found a ouabain-like action of metavanadate on active sodium transport. In addition, we report here a new biological effect of this compound on living cells: the inhibition of cyclic AMP-induced stimulation of transepithelial water flow.
