Summary
In cryostat-sections of mouse-kidney prefixed for 2–5 min in a solution of glutaraldehyde, the alkaline phosphatase activity after the CaCo-reaction and the CaPb-reaction is localized along the total cell surface. If the prefixing lasts 30 min, only the brush border is covered with the reaction product. These results must be attributed either to an inactivation of the enzyme by the fixing agent or — more likely — by partial dissolution of the enzyme (lyoenzyme ?) in the fixation fluid. The results obtained from cryostat-sections fixed for varying lengths of time are compared with findings from small blocks which were prefixed and in toto incubated, and with other small blocks, which were prefixed, cryostat sectioned and then incubated.
Zusammenfassung
In Kryostatschnitten von 2–5 min in Glutaraldehydlösung vorfixierten Mäusenieren ist die Aktivität der alkalischen Phosphatase sowohl nach der CaCo-Reaktion wie auch der CaPb-Reaktion über das gesamte Plasmalemm verteilt. Dauert die Vorfixierung 30 min, so wird nur der Bürstensaum dargestellt. Diese Ergebnisse sind entweder einer Inaktivierung des Enzyms durch das Vorfixierungsmittel zuzuschreiben oder sie sind, was wahrscheinlich gemacht wurde, durch Lösung eines Teils des Enzyms (Lyoenzym ?) im Fixierungsmittel bedingt. Die an verschieden lang fixierten Kryostatschnitten gewonnenen Resultate werden Befunden gegenübergestellt, die an in toto inkubierten, vorfixierten Blöckchen und an kyrostatschnittinkubierten. vorfixierten Blöckchen erhoben wurden.
