Summary
Voltage clamp experiments on single myelinated nerve fibres ofXenopus were done in which tetrodotoxin (TTX) and tetraethylammonium chloride (TEA) were applied to the excitable membrane from outside (by superfusion) or inside (by diffusion through the axoplasm from the cut end of the fibre).
The following results were obtained:

TTX (10−7 g/ml) in the external fluid selectively blocks the sodium current.

Even at a higher concentration TTX appears to have no effect from inside the membrane.

3.
0.3 mM TEA in the external fluid slows down the increase of the potassium current during a depolarizing pulse and reduces its steady state value.

4.
TEA applied from the cut end of the fibre to the inside of the nodal membrane also reduces the steady state potassium current but hastens its increase.

On the assumption that either side of the membrane is equally sensitive to TEA the diffusion coefficient for TEA ions in the axoplasm is obtained as 0.48×10−5 cm2sec−1 at 18° C. Control experiments gave a diffusion coefficient of 0.65×10−5 cm2sec−1 for sodium ions in the axoplasm.
The following conclusions were drawn: The properties of the sodium channel are different on the inner and outer surface of the nodal membrane. This agrees with similar findings on the squid axon membrane. The properties of the potassium channel are similar on either side. In this respect the nodal membrane differs from the squid axon membrane. There is no evidence that TTX or TEA can penetrate the nodal membrane.
Zusammenfassung
Einzelne Ranviersche Schnürringe des KrallenfroschesXenopus wurden mit der Technik des voltage clamp untersucht, um den Einfluß von Tetrodotoxin (TTX) und Tetraäthylammoniumchlorid (TEA) auf die Innen-und Außenseite der Schnürringsmembran zu studieren. TTX oder TEA gelangten an die Innenseite des untersuchten Schnürrings durch Diffusion im Axoplasma vom abgeschnittenen Ende der Nervenfaser her.
Es wurden folgende Befunde erhoben:

10−7 g/ml TTX blockieren bei Zusatz zur Außenlösung vollständig und selektiv die Natriumströme.

2.
An der Membraninnenseite scheint TTX selbst bei höherer Konzentration unwirksam zu sein.

3.
0,3 mM TEA verlangsamen bei Zusatz zur Außenlösung die Anstiegsgeschwindigkeit des Kaliumstromes und verkleinern dessen Maximalwert.

4.
Bei TEA-Einwirkung auf die Membraninnenseite wird ebenfalls der Maximalwert des Kaliumstromes verkleinert, die Anstiegsgeschwindigkeit des Kaliumstromes ist jedoch beschleunigt.

Unter der Annahme gleicher TEA-Empfindlichkeit des Kaliumstromes bei extracellulärer wie bei intracellulärer TEA-Einwirkung errechnet sich für TEA-Ionen bei 18° C ein Diffusionskoeffizient im Axoplasma vonD TEA=0,48·10−5 cm2 sec−1; Kontrollversuche ergaben für Natriumionen einD Na von 0,65·10−5 cm2sec−1.
Diese Befunde weisen darauf hin, daβ der Durchtrittsweg für Natriumionen offenbar an beiden Seiten der Schnürringsmembran verschieden aufgebaut ist. Den selben Schluβ lassen die entsprechenden Befunde anderer Autoren an der Riesennervenfaser des Tintenfisches zu. Der Durchtrittsweg für Kaliumionen verhält sich dagegen an beiden Seiten der Schnürringsmembran ähnlich. In dieser Beziehung unterscheiden sich die Membranen des Schnürrings und der Tintenfischfaser. Alle Befunde sprechen dafür, daβ die Schnürringsmembran undurchlässig für TTX und TEA ist.
