Summary
Phase contrast study of large bodies and protoplasts confirmed the findings obtained by staining methods specific for nuclear substance. Large bodies 3-4 hours old always contain more than two nucleoids (usually 3-5), while protoplasts usually have one (occasionally two). This is related to the manner of formation of the two formations and can be regarded as one of the features by which they can be distinguished.
The nucleoids of large bodies can fuse into a single central formation on media containing high salt or glucose concentrations. This effect is not abolished by ethylenediaminetetraacetic acid. Fusion does not occur on hypotonic media. Irradiation with u.v. light and cooling to 6-10ě C gave inconsistent results as far as fusion of the nucleoids was concerned. Fusion did not occur in large bodies fixed with osmium tetroxide or formaldehyde under the influence of a hypertonic medium. The hypothesis is expressed that fusion of the nucleoids is related to water loss (dehydration).
Gelatin was tested in 6-40% concentration as a medium for obtaining better contrast of the nucleoids, but with scant success.

лзучение крупных круглых телец (ККТ) и протопастов с помощью ϕазового контраста подтвтрдило данныт, полученные окраской специальными методами на ядерное нещество. ККТ в возрасте 3-4 часов содержат всетда больше, чем 2 нуклеоилы, -обычно 3-5, тогда как в протопастах наиболее часто находится 1 нуклеоид, в отдельных случаяс 2. Зто обусловлено способом нозникновения обоих обоих образований и, можно сказать, являтеся одним из их отличительных признаков.
Нуклеоиды ККТ могут сливаться в одно центральнре образование. Зто бывате в среде с повышенной концентрацитй солей или глюкозы. Зтилендиамонтетрау ксусная кислота на сннмает зтого дтйствия. Слияние не наблюдается в гипотонической среде. УФ облучение, как и охлаждение до 6-10оС не давали однозначных результатов, что касается слияния нуклеоидов. В ККТ, заϕиксированных осмиевой кислотой или ϕормальдегидом, слияние под действием гипертонической среды не наблюдается. Высказывается предположение, что слияние нуклеоидов связано с утратой воды (дтгиратацией).
В качтстве средства изображения нуклеоидов был испытан желатин в пределах от 6 до 40%, но с незначительным успехом.
