Summary
Between 1965 and the middle of 1969 24 strains ofListeria monocytogenes were cultured from 2873 human samples. This corresponds to an isolation rate of 0.87%. These findings were analysed with regard to the results of similar reports of other authors. They were also correlated to the age, sex, and clinical findings of the individuals from which positive cultures were obtained. The cold-enrichment merits particular attention in the attempt to isolate Listeria from various specimens. The isolation ofListeria monocytogenes from fecal cultures is rarely successful without the use of inhibitory substances in the culture media. Most of the tested inhibitory agents will not suppress the development of enterococci, the cultural properties of which render rather difficult the isolation ofListeria from the fecal samples.
In-vitro tests were carried out with mixtures ofL. m. and enterococci in 3,75% potassium-thiocyanate-broth (Lehnert) and promising results were obtained. The growth of enterococci was definitely suppressed during the first 4 days after inoculation while theListeria continued to multiply in this period. This fact was particularly noted after 48 hours of incubation at room temperature and subsequent storage at +4°C. 5Listeria monocytogenes strains cultured from 357 human specimens during the first half of 1969, were isolated by this method. The small number of positive results does not yet allow a final evaluation.
Zusammenfassung
Aus 2873 vom Menschen stammenden Untersuchungsproben wurde von 1965 bis Mitte 1969 24 malListeria monocytogenes (L. m.) kulturell nachgewiesen (0,87%). Diese Befunde wurden im Hinblick auf die Häufigkeit der Isolierungen im Verhältnis zu den Züchtungsergebnissen anderer Autoren in anderen Einzugsgebieten, auf Alter und Geschlecht der Probanden und deren klinischem Zustand analysiert. Besondere Beachtung verdient die Kälteanreicherung bei dem Nachweis von Listerien aus verschiedenen Untersuchungsmaterialien. Die Anzüchtung vonL. m. aus Stuhlproben gelingt nur selten ohne Anwendung von Substanzen, welche die Begleitkeime unterdrücken. Die meisten Hemmstoffe beeinträchtigen nicht das Wachstum von Enterokokken, die infolge ihrer kulturellen Eigenschaften die Isolierung von Listerien aus Stuhlproben am meisten erschweren.
Beiin vitro-Versuchen mit Keimgemischen aus Listerien und Enterokokken wurden günstige Ergebnisse mit 3,75% iger Kaliumthiocyanat-Bouillon (Lehnert) erzielt. Eine Unterdrückung des Enterokokkenwachstums war in den ersten 4 Tagen nach Einsaat unverkennbar. In diesem Zeitraum wurde ein Zunahme des Listerienwachstums beobachtet. Besonders ausgeprägt war dieser Effekt bei 48 stündiger Bebrütung bei Zimmertemperatur mit anschließender Aufbewahrung bei + 4°C. Mit dieser Methode wurden in der ersten Jahreshälfte 1969 in der Routinediagnostik 3 von 5 insgesamt gezüchtetenListeria- Stämmen aus 357 menschlichen Untersuchungsmaterialien isoliert. Für eine endgültige Beurteilung ist das Zahlenmaterial jedoch noch zu klein.
