Summary
Lymph was extracted from 8 healthy dogs by thoracic duct drainage and embedded for electron microscopic study, in a way explained before (Hebel u. Liebich, 1969). Small lymphocytes (88%), middle and great lymphocytes, plasmocytes and granulocytes were found in a proportion known from light-microscopic investigations. The morphological properties of cells were described and their size and nucleo-cytoplasmic ratio determined by planimetric measurement. The small lymphocytes of the dog's thoracic duct accord mostly with the results of the investigations of other animals in form and structure. Apart from the well known cell organelles and cell inclusions electron dense granulations were found in a small number of cells; they are interpreted as lysosomes. The meaning of smaller and more numerous granular or lamellated inclusions could not be explored. Centrioles could not be observed. The cell diameters of small lymphocytes come to 4.1Μ in average. The nuclear diameter of 3.0Μ in average works out a mean value of 1.51 for the nucleo-cytoplasmic ratio. The small lymphocytes show a nearly uniform cell population. The only indication for an eventual ageing — or specialising process — is the observation, that the cells with lower nucleo-cytoplasmic ratio show normally more cell organelles (mitochondria, Golgi-apparatus, ER) and have mostly a nucleolus, than those with high nucleo-cytoplasmic ratio.
Zusammenfassung
Von 8 klinisch gesunden Hunden wurde durch Drainage des Ductus thoracicus Lymphe gewonnen und nach einer früher beschriebenen Methode (Hebel u. Liebich, 1969) für die elektronenmikroskopische Präparation eingebettet. Etwa in dem aus lichtmikroskopischen Untersuchungen bekannten Verhältnis traten kleine Lymphocyten (88%), mittlere und gro\e Lymphocyten, Plasmocyten und granulocytäre Elemente auf. Die morphologischen Eigenschaften der Zellen werden beschrieben und durch planimetrische Verfahren in Grö\e und Kernplasmarelation ermittelt. Die kleinen Lymphocyten aus dem Ductus thoracicus des Hundes stimmen in Form und Bau weitgehend mit den Untersuchungsergebnissen an anderen Tierarten überein. Au\er den bekannten Zellorganellen und Zelleinschlüssen treten bei einer kleinen Anzahl von Zellen elektronenoptisch dichte Granula auf, die als Lysosomen gedeutet werden. Die Bedeutung von kleineren und wesentlich zahlreicher auftretenden, granulierten oder lamellären Körperchen konnte nicht geklärt werden. In keinem Schnittbild konnte ein Centriol beobachtet werden. Die Zelldurchmesser kleiner Lymphocyten liegen im Durchschnitt bei 4,1Μ. Bei einem durchschnittlichen Kerndurchmesser von 3,0Μ ergibt sich für die Kernplasmarelation ein Mittelwert von 1,51. Die kleinen Lymphocyten weisen ein nahezu uniformes Zellbild auf. Der einzige Hinweis auf einen eventuellen Alterungsbzw. Spezialisierungsproze\ besteht in der Beobachtung, da\ Zellen mit niedriger Kernplasmarelation in der Regel mehr Zellorganellen (Mitochondrien, Golgifeld, ER) aufweisen und häufiger einen Nucleolus besitzen als solche mit hohem Kernplasmaverhältnis.
