Summary
Cytomegalovirus infections are very dangerous for patients receiving an immunosuppressive therapy. Therefore, we investigated the influence of the immunsuppression, induced by cyclophosphamid onto the course of the infections withmurine cytomegalovirus in mice.
The disease is a suitable model for the investigation in animal experiments since many problems in human cytomegalo infections are unsolved due to the species specifity of the virus.
Virus-infected NMRI mice without cyclophosphamid pretreatment showed no symptoms of the disease, cytomegalichepatitis was minimal and short lasting, and the mortality was 1%.
All animals pretreated with cyclophosphamid and infected with the virus got sick, the mortality rate being high. After a single dose of 80 mg cyclophosphamid per kilogram body weight the mortality was 48%, after 3 doses of 80 mg/kg given prior to the infection, it was 84%. The infection of the liver lastet longer in all pretreated animals. The virus isolation was 70 times as high in animals given one dose of cyclophosphamid, and 570 times as high in those given 3 doses compared to animals without pretreatment.
Our experiments show clearly that even a single dose of cyclophosphamid is able to elevate the risk of infection during the immunsuppressive action, and that a relation exists between the effectivity of immunsuppression, mortality rate and virusmultiplication.
Zusammenfassung
Cytomegalievirusinfektionen stellen für Patienten, die unter einer immunosuppressiven Therapie stehen, eine ernste Gefahr dar.
Wir haben deshalb im Tierversuch mit Cyclophosphamid überprüft, welcher Zusammenhang zwischen der Stärke der Immunosuppression und dem Infektionsablauf besteht. Für die Versuche wurde das Mäusecytomegalievirus verwendet.
NMRI-Mäuse zeigen ohne Cyclophosphamidvorbehandlung nach der Virusinfektion keine Krankheitszeichen, die Mortalität ist gering (1%). Demgegenüber wiesen alle mit Cyclophosphamid vorbehandelte und infizierte Tiere schwere Krankheitszeichen auf. Die Mortalität betrug bei Tieren, die gleichzeitig mit der Virusverabreichung 1mal 80 mg/kg Cyclophosphamid bekamen, 48%, bei Tieren, die 3mal 80 mg/kg vor der Infizierung bekamen, 84%.
Die Virusausbeute in der Leber war bei einmaliger Cyclophosphamidvorbehandlung bis zu 70mal größer, bei dreimaliger Vorbehandlung bis zu 570mal größer als die in den nichtvorbehandelten Kontrolltieren.
Unsere Ergebnisse zeigen, daß die Verabreichung einer Einzeldosis von 80 mg/kg Cyclophosphamid die Empfänglichkeit von Mäusen gegenüber dem Mäusecytomegalievirus erhöht.
Eine Verstärkung der Immunosuppression zieht eine signifikante Erhöhung der Mortalität und der Virusmultiplikation in der Leber nach sich.
