Summary

A rapid, specific HPLC-method has been developed for quantitative determination of Ochratoxin A in food and feedstuffs as well as in mixed-feed.

After preliminary clean-up of the McOH/Chloroform extract the ochratoxin is analyzed using HPLC with a Spherisorb S5 ODS column McOH/water/acetic acid as eluent and a fluorescence detector.

The average recovery was about 50% in the range for 50–500 ppb with a small variation (coefficient of variation 10.24% and 23.650. The detection limit for Ochratoxin A is 5 ppb.

Zusammenfassung

Eine schnelle, spezifische HPLC-Methode zum quantitativen Nachweis von Ochratoxin A sowohl in Lebens- und Futtermitteln als auch im Mischfutter wird vorgestellt.

Ochratoxin A wird nach Vorreinigung im Schütteltrichter durch die mobile Phase Methanol/dest. Wasser (70+30)+1%. Essigsäure 100% von der Spherisorb S5 ODS-HPLC-Trennsäule eluiert und im Fluorescenzdetektor nachgewiesen.

Im Bereich von 50 und 500 ppb weisen die mittleren Auffindungsraten rund 50%, bei nur geringer Schwankungsbreite (Variationskoeffizient 10,24% und 23,650 auf. 5 ppb können eindeutig erfaßt werden.
