Effect of various oral contraceptives on the phagocytic activity of the reticulo-endothelial system

Summary

Two experiments were carried out on 480 female mice as well as on 40 female and 8 male test persons to study the influence of four oral contraceptives on the reticulo-endothelial phagocytic activity: Stediril-d®, Eugynon®, Lyndiol® and Noracyclin 22®.

The animals received the contraceptives suspended in a 1%-mucilage of tylose orally during their oestrus. The females took the pill regulary for 21 or 22 days, the males for 8 days.

The effect of the drugs on the phagocytic activity of the RES of the animals was measured by a carbon-clearance-test. All of the four contraceptives appearde to be significant in stimulating on the reticulo-endothelial phagocytic activity, after an initial depression. The activity from the beginning of the investigation was reached at least after 48 hours.

The phagocytic activity in man was measured by a lipofundin-clearance-test. Again a significant stimulation of the phagocytic activity of the RES was found in either sex without any difference.

It was shown with an additional experiment, that only the oestrogen-component of the contraceptives was responsible for the stimulation of the phagocytic activity of the RES. The progestagen-component was not able to stimulate the RES above a standard deviation.

Zusammenfassung

In 2 Versuchsreihen, an 480 weiblichen Mäusen sowie 40 weiblichen und 8 männlichen Versuchspersonen, wurde der Einfluß der vier Ovulationshemmer Stediril-d®, Eugynon®, Lyndiol® und Noracyclin 22® auf die RES-Phagozytose-Aktivität untersucht.

Dabei wurden den Tieren die Ovulationshemmer suspendiert in 1% igem Tyloseschleim per oral während ihres Östrus verabreicht. Die Frauen nahmen die Pille normal über 21 bzw. 22 Tage und die Männer 8 Tage lang.

Die Wirkung der Medikamente auf die Phagozytose-Aktivität des RES wurde bei den Tieren mit Hilfe des Carbon-Clearance-Testes festgestellt. Dabei zeigte sich, daß alle vier Ovulationshemmer nach einer initialen Depression eine signifikante Stimulation der RES-Phagozytose-Aktivität bewirkten, die erst nach 48 Std ihren Ausgangswert wieder erreichte.

Im Human-Versuch wurde die Phagozytose-Aktivität mit Hilfe des Lipofundin-Clearance-Testes bestimmt. Auch hier wurde, ohne Geschlechtsunterschied, eine deutliche Stimulation der Phagozytose-Aktivität des RES festgestellt.

In einem Zusatzversuch konnte nachgewiesen werden, daß lediglich die Östrogen-Komponente der Ovulationshemmer für die Stimulierung der Phagozytose-Aktivität des RES verantwortlich ist. Die Progestagen-Komponente vermochte das RES nicht über die Standardabweichung der Kontrolle hinaus zu stimulieren.
