Summary
1. Experiments on the isolated preparation of the guinea-pig aorta showed that the contraction caused by angiotensin II (50 ng/ml) is reduced to about 30% of the original height by cocaine (2mg/ml), phentolamine (3 μg/ml) and by repeated doses of tyramine (12 to 15 times 100 μg/ml).
2. The action of angiotensin on the isolated aorta of reserpine-pretreated (2 × 250 μg/kg i.m.) guinea-pigs is also diminished to the above mentioned degree.
3. The results suggest that about 2/3 of the angiotensin induced contraction of the aorta are mediated by the release of noradrenaline. The remaining direct action of angiotensin can not be influenced by additional doses of angiotensin (50 μg/ml).
4. After pretreatment of the guinea-pigs with α-methyldopa (3 × 400 mg/kg i.m.) more than 98% of the aortic catecholamines are α-methyl-noradrenaline. At this time the action of angiotensin is reduced to about 65%. Since α-methylnoradrenaline is 4 times less active than noradrenaline on this preparation, the result is consistent with the view that angiotensin releases also α-methylnoradrenaline.
5. On the aorta preparation of rats the action of angiotensin (50 ng/ml) is also reduced to about 30% by phentolamine (3 μg/ml).
6. Whereas on the isolated aorta of guinea-pigs angiotensin has lost its indirect activity after two doses, at least five doses of angiotensin are needed to reach the same result on the aorta of rats.
7. During suspension of the guinea-pig aorta in tyrode solution for two hours at 37∣dg C in the presence of tyramine (100 μg/ml) the noradrenaline content decreases to about 50%, whereas angiotensin in concentrations from 1 to 30 μg/ml is ineffective. From the isolated aorta of rats, however, angiotensin (1 μg/ml) releases 43% of the endogenous noradrenaline during the suspension period.
8. On the guinea-pigs blood pressure the normal potency of angiotensin relative to noradrenaline is 10, after pretreatment with α-methyldopa or reserpine the potency ratio is changed to about 5 or 3 respectively. The pressor effect of α-methylnoradrenaline on the blood pressure of rats and guinea-pigs is about 4 times less than that of noradrenaline.
9. The results demonstrate the existence of a direct and indirect mechanism of action of angiotensin on the isolated aorta of rats and guinea-pigs.
Zusammenfassung
1. Am isolierten Aortenpräparat des Meerschweinchens wird die durch Angiotensin ausgelöste Kontraktion nach Gabe von Cocain und Phentolamin sowie nach mehrmaliger Gabe von Tyramin auf etwa 1/5–2/5 ihrer ursprünglichen Höhe reduziert.
2. Auch nach Vorbehandlung mit Reserpin beobachtet man an der isolierten Aorta des Meerschweinchens eine Reduzierung der Angiotensinwirkung gleichen Umfanges.
3. Der nach Beseitigung der indirekten Komponente verbleibende Anteil der Angiotensinwirkung wird auch durch wiederholte Angiotensingabe nicht weiter reduziert.
4. Nach Vorbehandlung der Meerschweinchen mit 1-α-Methyl-Dopa ist die Angiotensinwirkung nur um etwa 1/3 vermindert. Da das aus α-Methyl-Dopa gebildete α-Methyl-Noradrenalin am Aortenpräparat etwa viermal schwächer wirksam ist als Noradrenalin, sprechen diese Ergebnisse dafür, daß Angiotensin auch α-Methyl-Noradrenalin freisetzt.
5. An der isolierten Rattenaorta wird die Angiotensinwirkung nach Phentolamingabe ebenfalls auf etwa 2/5 des ursprünglichen Effektes reduziert.
6. Während an der Meerschweinchenaorta nach zweimaliger Angiotensingabe die indirekte Wirkung aufgehoben ist, kann sie an der Rattenaorta erst nach mindestens fünf Angiotensindosen beseitigt werden.
7. Durch zweistündige Suspension in tyraminhaltiger Badflüssigkeit verarmt die Meerschweinchenaorta um 44% ihres Noradrenalingehaltes. Er wird jedoch durch Angiotensin nicht meßbar reduziert. An der Rattenaorta wird der Noradrenalingehalt demgegenüber auch durch Angiotensin um etwa 43% herabgesetzt.
8. Am Meerschweinchenblutdruck wird das Wirkverhältnis von Angiotensin zu Noradrenalin, das normalerweise 10:1 beträgt, nach Vorbehandlung mit 1-α-Methyl-Dopa auf etwa 5:1, mit Reserpin auf etwa 3:1 reduziert. 1-α-Methyl-Noradrenalin ist am Ratten- und Meerschweinchenblutdruck etwa viermal schwächer wirksam als Noradrenalin.
9. Der Mechanismus der Angiotensinwirkung am isolierten Aortenpräparat von Ratten und Meerschweinchen wird diskutiert.
