Summary
In order to determine amounts of glucose-6-phosphate dehydrogenase in parotid gland tissue relative to age, 20 rats (Sprague Dawley NIH/HAN) of different ages were killed and their parotid glands prepared. The amount of glucose-6-phosphate dehydrogenase in parotid glands decreases considerably with increasing age of the animals (Fig.2). However, after polyacrylamide slab gel electrophoresis, protein patterns of parotid gland tissues of both young and old rats are very similar (Fig.4). When determining with this electrophoresis protein containing glucose-6-phosphate dehydrogenase, it is found that equal amounts of protein yield lower enzymatic activities with increasing age (Fig.5). This might indicate that an aging organism forms increasing amounts of inactive or less active enzyme protein, this possibly being a significant biochemical process during aging which manifests itself pathologically by cell degeneration and finally, cell death.
Zusammenfassung
Um den altersabhängigen Gehalt an Glucose-6-phosphatDehydrogenase in Ohrspeicheldrüsengewebe bestimmen zu können, wurden 20 verschieden alte Laboratoriumsratten (Sprague Dawley NIH/HAN) getötet und deren Gl. parotis präpariert. Der Gehalt an Glucose-6-phosphat-Dehydrogenase in den Ohrspeicheldrüsen nimmt mit dem Alter der Tiere erheblich ab. Demgegenüber ist das Proteinmuster von Ohrspeicheldrüsengewebe in der Polyacrylamidplattenelektrophorese bei jungen und alten Ratten sehr ähnlich. Bestimmt man das Glucose-6-phosphat-Dehydrogenase-haltige Protein in dieser Elektrophorese, ergibt sich bei gleichem Proteingehalt eine mit dem Alter abnehmende Enzymaktivität. Das könnte bedeuten, daß der Organismus im Alter zunehmend inaktives oder weniger aktives Enzymprotein bildet, und dies ein wesentlicher biochemischer Vorgang im Alterungsprozeß ist, der sich pathologisch mit Zelldegeneration und Zelltod manifestiert.
