Plant regeneration from direct organogenesis of Pandanus canaranus Warb, an endemic medicinal plant

Abstract

Pandanus canaranus is an endemic medicinal plant that has been used for traditional medicines and many pharmaceutical applications. The current study is the first report for a development of efficient protocol for in vitro regeneration by direct or indirect organogenesis, which is necessary for the conservation of this important medicinal plant species. In the current study, we standardize the culture medium supplemented with different plant growth regulators (PGRs) of various concentrations using shoot tip, leaf, shoot and root meristematic explants. The shoot and root meristematic explants did not produce any callus whereas, leaf explants initiated callus combination with 1.5 mgl−1 of each 2ip + BAP PGRs. But, there was no further development and the callus was also browned in each and every subculture on same combination of the medium contains PGRs. After various PGRs and different concentration tested, cytokinin and auxins were influenced and showed multiple shoots formation from the shoot tip explants. The maximum number of multiple shoots and shoot length (elongation) were observed with 4 mgl−1 2ip and 3 mgl−1 of each BAP + GA3, respectively. Further, maximum number of root induction/development from elongated shoots were observed in the MS medium supplemented with 3 mgl−1 IBA. As per our knowledge and references, current study is the first report of successful development and establishment of in vitro regeneration in P. canaranus.
