Abstract
Polyneuropathy is defined as a simultaneous malfunction of several peripheral nerves, which could be a side effect of cancer therapy; however, this is reported to occur rarely and difficult to prove. The purpose of this study was to introduce at the first time the calorimetry in the diagnosis of neuropathy in an experimental animal model. Adult guinea pigs (n = 55, in 11 different groups) were injected intraperitoneally with the dose of cyclophosphamide that is comparable to the human dosage. Animals were euthanized; nerve (ischiadicus) and muscle (gastrocnemius) samples were analyzed by a SETARAM Micro-DSC. The denaturation temperatures as well as the calorimetric enthalpies were determined. The thermal parameters of denaturation of the samples varied depending on the therapeutic cyclophosphamide doses. The nerves were more sensitive to chemotherapy, compared to the muscles. An interesting body side effect of treatment was observed. The DSC is an appropriate tool to follow the effects of cyclophosphamide on peripheral nerves and muscles, which were found dose dependent.
