Summary
The antilipolytic compound 3, 5-dimethylisoxazole reduces the supply of energy yielding substrates in the fasting rat by lowering the concentration of unesterified fatty acids and ketone bodies in the blood. A rise in plasma corticosterone enables the animals to compensate for the loss of these important substrates by increasing glucose formation from aminoacids. Thus in fasting rats treated with dimethylisoxazole more 14C was incorporated from glycine into glucose. The serum urea level and the excretion of nitrogen in the urine was increased and liver tissue taken from such rats formed more glucose from alanine and incorporated more 14C from bicarbonate into glucose when incubated in vitro. As fed animals do not depend on lipolysis to maintain energy supply, plasma corticosterone levels, hepatic gluconeogenesis and serum urea levels were not increased by dimethylisoxazole in fed rats. When dimethylisoxazole was injected into fasting adrenal-ectomised rats hepatic gluconeogenesis was not stimulated. As adrenalectomised rats were thus unable to make up for the loss in fatty acids and ketone bodies by an additional formation of glucose from amino acids they succumb to severe hypoglycaemia. According to these results hepatic gluconeogenesis from amino acids is stimulated in fasting rats when lipolysis in adipose tissue is reduced by dimethylisoxazole. Our data are in agreement with the assumption that this effect depends on the action of endogenous corticosterone.
Zusammenfassung
Bei hungernden Ratten wird die Konzentration der als Energieträger wichtigen unveresterten Fettsäuren und Ketonkörper im Blut durch 3, 5-Dimethylisoxazol erniedrigt. Ein starker Anstieg des Plasma-Corticosteron setzt die Tiere instand, diesen Ausfall für die Energiestoffwechsel durch eine gesteigerte Gluconeogenese auszugleichen. Es wird vermehrt 14C aus Glycin in Glucose eingebaut, die Harnstoff konzentration im Serum und die Stickstoffausscheidung im Urin steigt an. Leberschnitte von den behandelten Tieren bilden in vitro mehr Glucose aus Alanin und bauen mehr 14C aus Bicarbonat in Glucose ein. Gefütterte Tiere sind nicht auf die Lipolyse angewiesen, um ihren Energiebedarf zu decken. Daher wurde bei gefütterten Ratten weder die Konzentration von Corticosteron im Plasma noch die Gluconeogenese oder die Harnstoffkonzentration im Serum durch Dimethylisoxazol gesteigert. Auch die Gluconeogenese im Lebergewebe von adrenalektomierten, hungernden Ratten wurde durch die Behandlung mit Dimethylisoxazol nicht erhöht. Da die adrenalektomierten Tiere den Verlust von Fettsäuren und Ketonkörpen nicht durch zusätzlich aus Aminosäuren gebildete Glucose ausgleichen können, geraten sie in schwerste, oft tödliche Hypoglykämien. Die Befunde zeigen, daß die Gluconeogenese in der Leber ansteigt, wenn hungernde Tiere daran gehindert werden, über ihre Fettdepots zu verfügen. Die Ergebnisse sprechen dafür, daß diese Reaktion durch endogenes Corticosteron ermöglicht wird.
