Abstract

ONO-3144 (2-aminomethyl-4-tert-butyl-6-propionylphenol hydrochloride) is a nonsteroidal antiinflammatory agent, which does not inhibit cyclo-oxygenase activity (1). The antiinflammatory action of this compound is thought to be due to its ability to scavenge proinflammatory free radicals (1–3). Prostaglandin (PG) biosynthesis is affected, however, by ONO-3144 stimulating PG hydroperoxidase activity converting PGG2 to PGH2, and inhibiting thromboxane synthetase. Despite the facilitated formation of PGH2 and consequently of PGE2, an important mediator of fever (4), ONO-3144 has been reported to produce antipyresis in febrile rats (5). In view of these apparently paradoxical effects by ONO-3144 on PGE2 production and fever in the rat, and because antipyretic drugs can have different effects in models of fever in rats and rabbits (6), the potential antipyretic activity of ONO-3144 in rabbits has been investigated in this study.
