Correlation between Antimicrobial Activity and Peroxidase Content of Leukocytes

Abstract

MICROBICIDAL assays on polymorphonuclear leukocytes (PMN) have been reported1–4, but their application as clinical tests has been delayed, partly because of the relatively large blood volumes required1–3 and the possible damage to the phagocytosing cells during the separation procedure3. Also, accurate determination of the viability of phagocytosed microbes is a problem. Previous methods involved either a disruption of the PMN or indirect criteria for viability such as the incorporation of 3H-thymidine4, the presence of buds on yeast cells3, or histochemical integrity of RNA3. Our work aimed to develop a direct and sensitive micro-scale method to distinguish small differences in the microbicidal activity of PMN and to evaluate the role of myelo-peroxidase (MPO) in this microbicidal function of PMN. The extent to which MPO is important for the intracellular killing5 remains questionable.
