Effects of the Method of Tissue Preparation on the Assay of Tissue Enzyme Activities

Abstract

THERE has recently been considerable interest in comparative enzyme-levels of various tissues of different species in health and disease. It is evident that the quantitative determination of enzyme activity in tissues is dependent not only upon the method used for measuring the activity, but also upon the method used for preparation of the tissue for analysis. Various factors, such as the intra-cellular localization of the enzyme, the availability of coenzymes and activators, and the permeability of both cellular and particulate membranes of different tissues to various substrate and product molecules, play a part in the effect of the preparation of the tissue prior to enzyme assay.
