Summary
In 6 male volunteers the rate of phenacetin elimination was not affected by treatment with phenobarbital.
In addition, the rate of phenazone elimination was determined in 3 of the 6 subjects. It increased by 40 per cent after treatment with phenobarbital.
The results demonstrate that hydroxylation of drugs by endoplasmic reticulum of liver cells is not equally enhanced by phenobarbital in man. Therefore, a drug, that stimulates enzyme production, does not necessarily increase the elimination rate of all the drugs metabolized by this enzyme system.
Zusammenfassung
An 6 freiwilligen Versuchspersonen wurde die Geschwindigkeit der Elimination von Phenacetin vor und nach Behandlung mit Phenobarbital bestimmt. Die Phenacetin-Elimination änderte sich durch die Behandlung mit Phenobarbital nicht.
An 3 der 6 Personen wurde zusätzlich die Geschwindigkeit der Elimination des Phenazins bestimmt; durch die Behandlung mit Phenobarbital stieg sie um 40% an.
Die Versuche zeigen, daß Phenobarbital nicht alle mikrosomalen Umsetzungen von Arzneimitteln im Menschen gleichmäßig steigert. Deshalb wird durch ein Arzneimittel, welches das hydroxylierende Enzymsystem der Leber stimuliert, nicht die Eliminationsgeschwindigkeit aller von diesem System oxydierten Medikamente beschleunigt.
