Zusammenfassung

Ein Alcianblau färbbares, in Perchlorsäure lösliches, antigenes saures Mucoprotein wurde aus menschlichem Siegelringzellkarzinom des Magens mit DEAE-Cellulose Chromatographie, sephadex G-200 und präparativer Polyacrylamid Gel Elektrophorese isoliert. Spezifische Immunseren wurden erzeugt, die in der indirekten Immunoenzym Histologie mit den Becherzellen des Dünn- und Dickdarms und mit den Becherzellen der metaplastischen Magenschleimhaut reagierten. In Resektionspräparaten des Magens (n=100) und des Colons (n=19) war in den 3 Siegelringzellkarzinomen des Magens und in den 3 Gallertkarzinomen des Dickdarms GOA nachweisbar. Hiermit wird eine immunchemische Beziehung von Becherzellen des Darmes mit diesen schleimbildenden Karzinomen von Magen und Colon demonstriert.

Summary

An Alcian blue staining, perchloric acid-soluble, antigenic acidic mucosubstance (GOA) was purified from human gastric signet ring cell carcinoma with DEAE-cellulose chromatography, Seqhadex G-200 and preparative polyacrylamide gel electrophoresis. Specific antisera were raised which reacted in indirect immunoenzyme histology with normal goblet cells of the small and large intestine and with goblet cells of intestinalized gastric mucosa. In surgical resection specimens of the stomach (n=100) and of the colon (n=19) 3 gastric signet ring cell carcinomas and 3 colonic colloidal adenocarcinomas stained for GOA, demonstrating an immunochemical relationship with normal intestinal goblet cells.
