Summary
Cell cultures of human lymphocytes from 20 healthy persons were stimulated to divide by phytohemagglutinin. Chromosome pulverization and mitotic inhibition occurred under the influence of 10−6 and 0,5×10−6 g sodium-diarsenate pro ml medium. In all experiments in control cultures without arsenic, these effects did not occur. According to a short review of the literature concerning similar results obtained with cytostatic drugs, DNA and RNA malformation, following inhibition of inbuilding of purine bases, is discussed as a possible cause of the damages. The possible value of the results in relation to the carcinogenic effect of anorganic arsenic compounds is discussed.
Zusammenfassung
Lymphocytenkulturen von 20 gesunden Spendern wurden durch Phytohaemagglutinin zu mitotischer Aktivität angeregt. Unter der Einwirkung von 10−6 und 0,5·10−6 Natriumbiarsenat pro ml Medium fanden wir in allen Versuchsansätzen Chromosomenpulverisation und Hemmung der mitotischen Aktivität. In den entsprechenden Kontrollkulturen wurden diese Effekte nicht beobachtet. Unter Bezug auf vergleichbare Ergebnisse mit Cytostatica und deren Deutung wird als Ursache dieser Veränderungen eine DNS- und RNS-Fehlbildung infolge Störung des Purinbaseneinbaus diskutiert. Ob diesem Befund eine mögliche Bedeutung im Hinblick auf die carcinogene Wirkung anorganischer Arsenverbindungen zukommt wird erörtert.
