Characterization ofPhytophthora infestans from Maine during 1999 and 2000

Abstract

A total of 77 isolates ofPhytophthora infestans was collected in 1999 and 93 isolates were collected in 2000 from growers’ fields, home gardens, and research plots in Maine. Genotype diversity based onglucose-6-phosphate isomerase (Gpi) allozyme analysis was greater in the collections from 2000, with six genotypes identified vs two genotypes identified in 1999. The US-8 (Gpi 100/111/122) genotype constituted 88% of the isolates collected in 1999 and 75% of those collected in 2000. The US-14 (Gpi 100/122) genotype constituted 12% and 7% of the isolates collected in 1999 and 2000, respectively. Three newGpi genotypes constituted 7% of the isolates collected from potato in northern Maine in 2000. A new genotype (A1 mating type,Gpi 100/100) that was aggressive on both tomato and potato was identified from home gardens in western Maine and constituted 11% of the isolates collected in 2000. A1, A2, and A1A2 mating types as well as self-fertile isolates were collected in both years. Self-fertility in artificial culture appeared to be relatively common in field isolatesof P. infestans. Potential origins of the new genotypes are discussed.

Resumen

Un total de 77 aislamientos deP. infestans se recolectaron en 1999 y otros 93 fueron recolectados en el 2000 en campos de cultivo, huertos familiares y parcelas de investigación de Maine. La diversidad genotípica basada en el análisis de aloenzimas deglucosa-6-fosfato de isomerasa (Gpi) fue mayor en las colecciones del 2000 con seis genotipos identificados versus dos genotipos identificados en las de 1999. El genotipo US-8 (Gpi 100/111/122) comprendió al 88% de los aislamientos colectados en 1999 y al 75% de los colectados en el 2000. El genotipo US-14 (Gpi 100/122) comprendió 12% y 7% de los aislamientos colectados en 1999 y 2000, respectivamente. Tres nuevos genotiposGpi abarcaron al 7% de los aislamientos colectados de las papas del norte de Maine en el 2000. Un nuevo genotipo (grupo de apareamiento A1,Gpi 100/100) de comportamiento agresivo tanto en el tomate como en la papa fue identificado en los huertos familiares del oeste de Maine y comprendió al 11% de los aislamientos colectados en el 2000.
