Abstract
In concentrations higher by one order of magnitude than is the optimal concentration for stimulating chlortetracycline (CTC) biosynthesis, benzyl thiocyanate inhibits the oxidation of sucrose by the washed mycelium ofStreptomyces aureofaciens. At 6×10−4M concentration, it inhibits the rate of sucrose oxidation by 76%. Under these conditions the oxidation of organic acids and amino-acids and endogenous respiration are not inhibited. Endogenous respiration begins to be blocked at higher concentrations of benzyl thiocyanate. Benzyl thiocyanate is most effective when added before sucrose. When it is added simultaneously with or after sucrose its effect is considrably lower. The blocking effect of benzyl thiocyanate can be removed by washing the mycelium with water. Benzyl thiocyanate added to a mycelium suspension before substrate acts as a noncompetitive inhibitor of sucrose oxidation. The hypothesis is advanced that benzyl thiocyanate acts as inhibitor of sucrose transport into the mycelium.
Abstract
Вконцентрациях, превышающих оптимальную концентрацию, которая стимулирует биосинтез CTC, роданистый бензил угнетает окисление сахарозы промытым мицелием Streptomyces aureofaciens. В концентрации 6. 10−4 M он понижает скорость окисления сахарозы на 76 %. При этих условиях еще не угнетаются ни окисление органических кислот и аминокислот, ни эндогенное дыхание. Угнетение эндогенного дыхания начинается только при более высоких концентрациях роданистого бензила. Наиболее резкое ингибиторное действие роданистого бензила на окисление сахарозы наблюдается при его прибавлении перед прибавлением субстрата. При прибавлении после сахарозы или даже одновременно с ней действие роданистого бензила бывает гораздо ниже. Угнетающее действие роданистого бензила на окисление сахарозы снимается при промывании мицелия водой. Роданистый бензил, прибавленный к суспензии мицелия еще до прибавления субстрата, действует как некомпетитивный ингибитор окисления сахарозы.—Высказывается предположение, что роданистый бензил действует как ингибитор транспорта сахарозы в мицелий.
