Summary
New results in the application of TLC to the analysis of amino acids in biological material, as their Dinitrophenyl (DNP)-derivatives, are reported.
The non-destructive detection of amino acids using Dinitrofluorobenzene (DNFB) allows the reaction-chromatography of these compounds, i.e.:

a)
in situ synthesis of DNP-derivatives on the layer, followed byone-two-dimensional chromatography.

b)
one-dimensional separation offree amino acids, conversion with DNFB and chromatography of DNP-compounds in thesecond dimension. Since the amino acids are not destroyed after detection with DNFB, these techniques make possible moreunambiguous identification by means ofmulti-dimensional techniques.
Nucleic acid bases, nucleosides and nucleotides can be separated by means of a new transfer technique (Autotransfer TLC), consisting of transfer ofnucleic acid bases andnucleosides frompolyethylenimine (PEI)-cellulose to cellulose and separation of these compounds on cellulose layers, whereas thenucleotides are separated onPEI-cellulose layers.

Neuere Ergebnisse in der Anwendung der DC zur Analyse von Aminosäuren in biologischem Material, in der Form ihrer DNP-Derivate, werden beschrieben.
Der zerstörungsfreie Nachweis von Aminosäuren mittels DNFB erlaubt die Reaktions-Chromatographie dieser Verbindungen, d. h.:
-
a)
in situ Synthese von DNP-Derivaten auf der Schicht, gefolgt von ein- oder zweidimensionaler Chromatographie.

b)
eindimensionale Trennung der freien Aminosäuren, Umsetzung mit DNFB, und Chromatographie der DNP-Verbindungen in der zweiten Dimension. Da die Aminosäuren, nach dem Nachweis mittels DNFB unzersetzt vorliegen, erlauben diese Techniken eine eindeutige Identifikation, z.B. Anwendung der mehrdimensionalen Technik.
Nucleinsäure Basen, Nucleoside und Nucleotide lassen sich mit Hilfe einer neuen Transfer-Technik (Autotransfer DC) trennen. Nucleinsäure Basen und Nucleoside werden von PEI-Cellulose auf Cellulose transferiert, und auf Cellulose-Schichten getrennt, während die Trennung von Nucleotiden auf PEI-Cellulose-Schichten erfolgt.

On décrit des résultats récents obtenus par la CCM, employée pour l'analyse d'amio-acides dans des matières biologique sous forme de leurs dérivés DNP.
La détection sans destruction d'amino-acides au moyen de DNFB permet d'effectuer la chromatographie de réaction sur ces composés, c'est-à-dire:
-
a)
synthèse in situ des dérivés DNP sur la couche, suivie par chromatographie uni- ou bi-directionelle;
-
b)
séparation uni-directionnelle des amino-acides libres, réaction avec DNFB, et chromatographie des composés DNP en deuxième direction. Puisque les aminoacides, après détection au moyen de DNFB, sont présents sous forme non-décomposée, ces techniques permettent une identification non équivoque, par exemple l'application de la technique multi-directionnelle.
Des bases d'acides nucléiques, des nucléosides, et des nucléotides peuvent être séparés à l'aide d'une nouvelle technique de transfert (auto-transfert CCM). Les bases d'acides nucléiques et les nucléosides sont transférés de cellulose PEI sur des couches de cellulose où ils sont séparés, tandis que la séparation des nucléotides est effectuée sur des couches de cellulose PEI.
