Abstract

P. aeruginosa flagella of the complex serotype a (a0, a2, a3) and the uniform serotype b were isolated, purified, and depolymerized by heat. The flagellins obtained form one precipitin band each and show reactions of nonidentity in Ouchterlony tests. The immunologic reaction is not altered by treatment with SDS and mercaptoethanol. In disc microelectrophoresis and SDS gradient gel microelectrophoresis on polyacrylamide gels, the flagellins migrate as homogeneous proteins. They have identical electrophoretic mobilities and a molecular weight of about 50,000.

Zusammenfassung

P. aeruginosa-Flzgellen des komplexen Serotyps a (a0, a2, a3) und des uniform Serotyps b werden isoliert, gereinigt und durch Hitzebehandlung depolymerisiert. Die erhaltenen Flagelline zeigen in Ouchterlony-Tests jeweils eine einzige Präzipitatbande und Antigenreaktionen der Nicht-Identität. Die immunologische Reaktion wird durch Behandlung mit SDS und Mercaptoaethanol nicht beeinflußt. In der Disk-Mikroelektrophorese und SDS-Gradientengel-Mikroelektrophorese an Polyacrylamidgelen wandern die Flagelline wie homogene Proteine. Sie besitzen die gleiche elektrophoretische Mobilität. Ihr Molekulargewicht liegt bei 50 000.
