Summary
The DNA of the following herpesviruses was characterized for the first time by calculating the G+C content from the buoyant density in CsCl which was determined in the analytical ultracentrifuge: Varicella-Zoster virus, Cytomegaloviruses of African Green Monkeys, human Cytomegaloviruses, Equine Coitale Exanthema virus, Tree Shrew Herpesvirus and IBR/IPV viruses. The buoyant density and melting temperature of purified DNA from HSV-1, HSV-2, IBR- and Psr-virus was measured by biophysical methods and the G+C content could be calculated. The base composition of the DNA was chemically analyzed. From these four viruses and from V-Z and ECE virus the DNA-double-strandedness could be proved. The sedimentation coefficient of DNA from HSV-1, HSV-2 and Psr-virus was measured by analytical zone centrifugation and the molecular weight was calculated.
Zusammenfassung
Mit Hilfe der DNS-Flotationsdichtebestimmung in CsCl durch analytische Ultrazentrifugation konnten erstmalig folgende Herpesvirusstämme im G+C-Gehalt ihrer DNS charakterisiert werden: Varicella-Zoster Virus, Affen-Cytomegaloviren, Cytomegaloviren des Menschen, Equine Coitale Exanthema Virus, Tree Shrew Virus und IBR/IPV-Viren. An den gereinigten Nucleinsäuren des HSV-1, HSV-2, IBR- und Psr-Virus konnten die Dichte und der Schmelzpunkt biophysikalisch bestimmt und der G+C-Gehalt errechnet werden. Durch chemische Analyse wurde die Basenzusammensetzung ermittelt. Die Doppel-strängigkeit konnte bei der DNS dieser Viren sowie des V-Z-Virus und ECE-Virus nachgewiesen werden. Mit der analytischen Bandzentrifugationsmethode ließen sich für die DNS des HSV-1, HSV-2 und Par-Virus die Sedimentationskoeffizienten bestimmen und das Molekulargewicht berechnen.
