Summary
PP12 was isolated from aqueous extracts of human term placentae by fractionating the proteins with rivanol and ammonium sulfate, gel filtration, ion-exchange chromatography and the use of immunoadsorbents.
An average of 5 mg PP12 could be extracted from a human term placenta. In extracts from other human tissues this protein could not be detected; PP12 therefore appears to be a protein specific for the placenta.
In cord blood sera and in sera of pregnant women PP12 was absent or present only in trace amounts (< 0.1 mg/100 ml). However, PP12 occured in higher concentrations in amniotic fluid; the maximal values being found between the 20th and 30th week of pregnancy with an average of 11 mg/100 ml.
PP12 had the electrophoretic mobility of an α1-globulin and an isoelectric point of 4.6–4.7. The purified protein sedimented with 2.7 S. PP12 was found to have a molecular weight of 25,200 as determined by ultracentrifugation, but a molecular weight of 51,000 as determined by SDS-PAA gel electrophoresis.
PP12 is a glycoprotein and contains 4.3% carbohydrates (hexoses 2.8%, hexosamines 0.7%, fucose 0.03%, sialic acid 0.8%). The amino acid composition of PP12 has also been determined. The most abundant amino acids in this protein are glutamic acid, alanine, serine, and proline. p ]The molecules of PP12 are heterogeneous in size and charge. On immunizing animals with PP12 antisera binding with two quite different antigenic determinants were obtained. Proteins with a close immunochemical relation to PP12 occurred in very high concentrations in the placentae of rhesus (cynomolgus) monkeys.

PP12 wurde aus dem wäßrigen Extrakt reifer menschlicher Plazenten isoliert; die Reinigung erfolgte durch fraktionierte Fällung der Proteine mit Rivanol und Ammoniumsulfat, durch Gelfiltration, Ionenaustauschchromatographie und unter Verwendung von Immunadsorbentien. Aus einer ausgewachsenen menschlichen Plazenta lassen sich im Durchschnitt 5 mg PP12 extrahieren. Im Extrakt von anderen menschlichen Organen konnte dieses Protein nicht nachgewiesen werden; PP12 ist demnach offensichtlich spezifisch für die Plazenta. In Nabelschnurseren und in Seren schwangerer Frauen kommt PP12 normalerweise nicht oder nur in Spuren (< 0,1 mg/100 ml) vor. In größerer Konzentration wird PP12 im Fruchtwasser gefunden; die höchsten Werte werden dabei zwischen der 20. und 30. Woche der Schwangerschaft mit durchschnittlich 11 mg/100 ml gefunden.
PP12 hat die elektrophoretische Beweglichkeit eines α1-Globulins und einen isoelektrischen Punkt von 4,6–4,7. Das gereinigte Protein sedimentiert mit 2,7 S. In der Ultrazentrifuge wurde für PP12 ein Molekulargewicht von 25 200, im SDS- haltigen Polyacrylamidgel dagegen ein Molekulargewicht von 51 000 ermittelt.
PP12 ist ein Glykoprotein und enthält 4,3% Kohlenhydrate (Hexosen 2,8%, Hexosamine 0,7%, Fucose 0,03%, Neuraminsäure 0,8%). Die Aminosäurenzusammensetzung des Proteins wurde ebenfalls ermittelt; die in der Peptidkette am häufigsten Aminosäuren sind Glutaminsäure, Alanin, Serin und Prolin.
PP12 ist ein hinsichtlich der Größe und Ladung der Moleküle heterogenes Protein. Beim Immunisieren von Tieren mit PP12 werden meist Antikörper gegen 2 recht unterschiedliche Antigendeterminanten erhalten. Mit PP12 immunologisch nah verwandte Proteine kommen in sehr hoher Konzentration in Plazenten von Affen (Rhesus und Cynomolgus) vor.
