Summary
Isolated dog kidneys were perfused with blood in a recirculating pump-oxygenator system. After addition of albumin-bound C14-palmitate, the CO2-production, the plasma nonesterified fatty acids (NEFA) and the specific activities of CO2 and NEFA were determined over a period of 30–60 min. The mean NEFA uptake by the kidney was 1,5 μMol/min/100 g. By direct determination, the NEFA oxydation to CO2 averaged 0,43 μMol/min/100 g. As a result of the relatively short duration of the experiments, this figure must be considered as a minimum value. The true NEFA oxydation was calculated from the experimental data and averaged 1,12 μMol/min/100 g. According to this calculation, 7,5% of the CO2 formed by the kidney was derived from NEFA.
Zusammenfassung
Isolierte Hundenieren wurden in einem künstlichen Kreislauf mit Blut rezirkulierend durchströmt. Nach Zugabe von C14-Palmitinsäure wurde die Konzentration von unveresterten Fettsäuren (UFS) im Plasma, die CO2-Produktion und die spezifische Aktivität der UFS und des abgegebenen CO2 über 60 min gemessen. Im Mittel ergab sich eine UFS-Aufnahme von 1,5 μMol/min/100 g Niere. Für die vollständige Oxydation dieser Fettsäuremenge würden 10,2% der gleichzeitig gemessenen CO2-Produktion und 14,7% der O2-Aufnahme benötigt werden. Die direkte Messung der UFS-Oxydation ergab einen Wert von im Mittel 0,43 μMol/min/100 g Niere. Dieser Wert ist wegen der relativ kurzen Versuchsdauer nur als Mindestwert anzusehen. Die wahre Größe der UFS-Oxydation wurde aus den Versuchsdaten indirekt berechnet und betrug im Mittel 1,12 μMol/min/100 g Niere. Demnach entfielen im Mittel 7,5% des von der Niere gebildeten CO2 und 11,5% des renalen O2-Verbrauchs auf die Oxydation von UFS.
Der Anteil an der Energiebereitstellung durch Oxydation von unveresterten Fettsäuren wird im Vergleich mit dem Umsatz anderer Metaboliten in der Niere diskutiert.
