Summary
Procedures for preparing isolated islets and completely dissociated single islet cells free from the surrounding acinar tissue are described. The mixture of Ficoll and Conray solution (Ficoll-Conray solution) employed for the collection of islets is less viscous and toxic than the plain Ficoll solution and is also applicable for the separation of cells other than islets. Dissociation of islet cells by Dispase results in a satisfactory yield of single islet cells with viability greater than 95%. The islets and dispersed islet cells can be maintained in culture for several months with no deterioration of hormone-producing ability.
