Bestimmung der Aminosäuren und biogenen Amine in der dunklen und hellen Muskulatur des Thunfisches mittels Aminosäureanalysator und Hochdruckflüssigkeitschromatographie

Summary

The results of storage of the light and dark muscles of tuna are given and the methods used for determination of the biogenic amines and amino acids (HPLC, amino acid analysis), are described and discussed.

The conjecture that particularly the dark muscles contain large amounts of free histidine is not confirmed. There is no histamine detectable in the dark muscles even when stored at 30 °C for 40 days but high concentrations of cadaverine and putrescine are found within 5 days.

In contrast it histamine concentrations as high as 4,500 mg/kg are detected in the light muscles upon storage at 30 °C.

Zusammenfassung

Es werden die Ergebnisse von Lagerungsversuchen mit der hellen and dunklen Muskulatur des Thunfischs dargestellt, sowie die für die Bestimmung der Aminosäuren and biogenen Amine angewandten Verfahren (Aminosäureanalyse, HPLC) beschrieben und diskutiert.

Die bis in die jüngere Literatur vertretene Ansicht, daß besonders die dunklen Muskelpartien reich an freiem Histidin und somit als potentielle Histaminbildner von Bedeutung sind, wird nicht bestätigt. In der dunklen Muskulatur kann auch nach 40tägiger Lagerung bei +30 °C kein Histamin nachgewiesen werden, während bereits nach 5 Tagen sehr hohe Cadaverin- and Putrescingehalte gefunden werden. Dagegen werden in der hellen Muskulatur bei 30 °C Histamin-Konzentrationen bis zu 4 500 mg/kg erreicht.
