Summary
In an investigation, in the rat, of the effect of intracerebrally injected emulsions of sphingolipids extracted from normal human brains and from human cases of metachromatic leucodystrophy and of Krabbe's disease the local reactions of the cerebral tissue were studied at varying intervals after the injection.
The tissue reaction to the injection of cerebroside emulsions was characterised, as in human cases of Krabbe's disease, by the occurrence of “globoid cells” and “globoid bodies,” while injection of sulphatide emulsions was followed by the occurrence of substances staining metachromatically brown with cresyl violet-acetic acid and showing only a slight tendency to undergo phagocytosis as in human cases of metachromatic leucodystrophy particularly those with short duration.
These observations lend support to the assumption that the difference between the histopathological picture in metachromatic leucodystrophy and Krabbe's disease is due to the chemical properties of the accumulated substances.
Zusammenfassung
In einer Untersuchung an der Ratte über die Wirkung von intracerebral injizierten Sphingolipoidemulsionen, die aus normalen menschlichen Gehirnen sowie von Humanfällen metachromatischer Leukodystrophie und Krabbescher Erkrankung extrahiert worden waren, wurden die lokalen Reaktionen des Hirngewebes in verschiedenen Zeitabständen nach der Injektion untersucht.
Die Gewebsreaktion auf Injektion von Cerebrosid-Emulsionen war, wie bei den Humanfällen von Krabbescher Erkrankung, durch das Auftreten von „Globoidzellen” und „Globoidkörpern” charakterisiert, während nach Injektion von Sulphatid-Emulsionen Substanzen auftraten, die bei Färbung mit essigsaurem Kresylviolett eine Braun-Metachromasie zeigen und nur geringe Phagocytosetendenz zeigen, ähnlich wie bei menschlichen Fällen von metachromatischer Leukodystrophie, besonders bei denen von kurzer Dauer.
Diese Beobachtungen stützen die Annahme, daß die Unterschiede im histopathologischen Bild der metachromatischen Leukodystrophie und der Krabbeschen Erkrankung auf die chemischen Eigenschaften der angehäuften Substanzen zurückzuführen sind.
