Zusammenfassung
Es wird über die erfolgreiche Züchtung und vergleichende enzymhistochemische Untersuchung eines Opticusspongioblastoms berichtet.
Im frisch eingefrorenen Material enthielten die größtenteils spindelförmigen Zellen hohe Aktivitäten von DPN-Diaphorase und Lactatdehydrogenase. Die Fermente des Citronensäurecyclus waren nur schwach oder wie die Succinodehydrogenase nicht nachweisbar. Die gleichen enzymhistochemischen Befunde wurden an den Gewebekulturen erhoben. Das Wachstum des “Opticusglioms” entsprach in allem den vonKersting gegebenen Kriterien für das Spongioblastom des Kleinhirns. Wir möchten uns deshalb der Meinung vonZülch u.a. anschließen und den Tumor den Spongioblastomen zuordnen.
Im Verlaufe der Züchtung gelang es, das Auftreten von Rosenthalschen Fasern zu verfolgen. Sie ließen sich als Degenerationsprodukte der Geschwulstzellen identifizieren und enthielten keinerlei oxydative Enzyme.
Summary
An optic nerve glioma was successfully grown in tissue culture and morphological and enzyme-histochemical studies of this tumor were carried out over a period of 3 months. During this interval the tissue-culture cells maintained a homogenous spindle-shaped bipolar appearance and no development of mature glial cells was seen. The morphological similarity of these tissue culture cells to cerebellar astrocytoma tissue-culture cells described byKersting led us to classify this tumor as a spongioblastoma, according toZülch.
Enzyme-histochemical studies were undertaken on the original surgical tumor specimen as well as on the tissue culture cells. The histochemical reactions in the original tumor and in periodic samplings of the tissue-culture cells were always consistent. The cells showed a very strong activity of DPN-diaphorase and lactat dehydrogenase (no activity of succinodehydrogenase), and a minimal activity of the other enzymes in the citric acid cycle.
During the tissue-culturing process the development ofVerhoeff's “cytoid bodies” (Rosenthalsche Fasern) was observed. Because of the frequent observation of these “cytoid bodies” within degenerating tumor cells and the absence of enzymatic activity within these structures these “cytoid bodies” were felt to represent degenerating products of tumor cells.
