Summary
The influence of external calcium and sodium ions1 on potassium contractures was studied in different isolated preparations of, mammalian cardiac muscle (thin ventricular and atrial trabeculae of calf hearts; guinea-pig papillary muscles).
In atrial trabeculae of calf hearts, the effects of external calcium and sodium ions on contractures were the same as in guinea-pig auricles (Scholz 1969a). Raising [Ca] e or reducing [Na] e caused an increase of tension development and a shift of mechanical threshold to lower [K] e . At any given [K] e the amplitude of contractures depended on the ratio Ca/Na2 in the bathing fluid as in frog hearts (Lüttgau andNiedergerke, 1958). In Na-free solutions contractures developed even without increasing [K] e .
Compared to the atrial preparations, however, contractures of ventricular trabeculae of calf hearts and of guinea-pig papillary muscles showed the following differences. Raising [Ca] e or reducing [Na] e caused a somewhat smaller increase in tension development. In Na-free solutions, no contractures could be observed unless [K] e was increased. At constant [K] e , the amplitude of contractures did not depend on the ratio [Ca] e /[Na] e 2.
The differences between the mechanical response of atrial and ventricular preparations of mammalian hearts are tentatively explained by some differences in the ultrastructure of both tissues.
Zusammenfassung
An isolierten Präparaten aus Säuparaten aus, Säugetierherzen (dünne Trabekel aus Vorhof und Ventrikel von Kalbsherzen; Meerschweinchenpapillarmuskeln) wurde der Einfluß von Calcium- und Natriumionen auf das Verhalten der Kaliumkontraktur untersucht.
Kontrakturen von Kalbsvorhofstrabekeln glichen in ihrem Verlauf und ihrer Beeinflußbarkeit durch Ca und Na vollständig denen des Meerschweinchenvorhofs (Scholz, 1969a). Eine Steigerung der [Ca] e oder eine Erniedrigung der [Na] e bewirkte eine Verstärkung der Kontrakturen und eine Verschiebung der Kontrakturschwelle zu niedrigeren [K] e . Bei gegebener [K] e wurde die Kontrakturhöhe bestimmt durch das Ionenverhältnis Ca/Na2 in der Badlösung. In Na-freier Lösung kam es auch ohne Erhöhung der [K] e zur sogenannten Na-Entzugskontraktur.
Kontrakturen von ventrikulären Kalbstrabekeln und Meerschweinchenpapillarmuskeln zeigten gegenüber denen von Vorhofspräparaten aus Warmblüterherzen im wesentlichen folgende Unterschiede. Sie wurden durch Erhöhung der [Ca] e oder Erniedrigung der [Na] e zwar ebenfalls verstärkt waren aber insgesamt gesehen unempfindlicher gegenüber Änderungen der extracellulären Calcium-bzw. Natriumkonzentration. Auch in Na-freier Lösung mußte die [K] e immer erhöht werden, um Kontrakturen auszulösen, d.h., es kam nicht zur Na-Entzugskontraktur. Bei gegebener [K] e folgten sie nicht dem Verhältnis [Ca] e /[Na] e 2.
Das unterschiedliche Kontrakturverhalten bei Vorhofs- und Ventrikelpräparaten von Warmblüterherzen wird versuchsweise erklärt durch Unterschiede in der Ultrastruktur (transversales Tubulussystem und sarcoplasmatisches Reticulum) bei beiden Gewebsarten.
