Summary
Guinea pigs were treated with subtoxic or with toxic doses of diisopropyl-fluoro-phosphate (DFP), which was administered intravenously. Smaller amounts of the drug (0.1 mg/kg) are preferably bound by the serum but also by the lungs. After the administration of higher doses (3 and 6 mg/kg) an uniform distribution of the drug over the body is observed, whereas an accumulation occurs in the liver and the kidney. An important part of the injected DFP is metabolized in the organism, to yield diisopropyl-phosphate (DP). The excretion of DFP and DP mainly occurs with the urine and to a smaller extent with the bile. A small part of the injected DFP is excreted as such. The greater part of it, however, is excreted as DP. Upon administration of highly toxic doses the urinary excretion is reduced, whereas the concentration of DFP and its metabolites in the bile increases.
The Rf value of3H-DFP on a thin-layer radiochromatogram proves 0.38 and that of3H-DP 0.27.
Zusammenfassung
Diisopropyl-fluoro-phosphat (DFP) wurde Meerschweinchen in subtoxischen und toxischen Dosen intravenös injiziert. Niedrige Konzentrationen (0,1 mg/kg) werden vor allem im Serum, aber auch in der Lunge gebunden, bei höheren Dosen (3 und 6 mg/kg) verteilt sich die Substanz nahezu gleichmäßig über den Organismus und reichert sich nur in Leber und Niere an. Ein wesentlicher Teil von DFP wird im Organismus zu Diisopropyl-phosphat (DP) abgebaut. Die Ausscheidung von DFP und DP erfolgt hauptsächlich im Urin, in geringem Ausmaß mit der Galle. DFP wird in kleinen Mengen unverändert, in der Hauptsache aber als DP ausgeschieden. Bei Applikation hochtoxischer Dosen geht die Urinausscheidung zurück, während der Gehalt an DFP und seinen Stoffwechselprodukten in der Galle ansteigt.
Die Radio-Dünnschichtchromatographie ergibt für3H-DFP einen Rf-Wert von 0,38, für3H-DP von 0,27.
