Summary
Pre- and postchromatographic derivatization reactions are used in quantitative TLC in order to enhance the specifity and sensitivity of the analytical procedures.
The introduction of chromophores and fluorophores enables the quantitative analysis in the ng-pg/spot range.
Estriol is derivatized to azo-dyes and dansyl esters; selenium is determined as 2,1,3-benzoselenodiazole or fluorescing 2,1,3-naphthoselenodiazole. Besides the direct determination of Cortisol the condensation to Cortisoldansylhydrazone is described. Fluorescing testosterone derivates are derivatized by postchromatographic reaction with (a) isonicotinic acid hydrazide, (b) on basic Al2O3-plates and (c) with ammonia hydrogencarbonate, or by prechromatographic derivatization with dansylhydrazone. Lecithin and sphingomyelin are derivatized with MnCl2 to fluorescing compounds and carbamazepin by treating with HCl-fumes or with HClO4-solutions. Volatile carboxylic acids are derivatized to p-bromo-phenacyl esters, naphthacyl esters or DAPACAP esters.
Zusammenfassung
Prä- und postchromatographische Derivatisierungsreaktionen werden in der quantitativen Dünnschicht-Chromatographie zur Steigerung der Spezifität und Empfindlichkeit der Analysenverfahren eingesetzt.
Neben der Einführung von Chromophoren in das Molekül werden vor allem Fluorescenzreaktionen beschrieben, die eine quantitative Bestimmung im pg/Fleck-Bereich ermöglichen.
Östriol wird zu Azofarbstoffen und Dansylestern derivatisiert; Selen zu 2,1,3-Benzoselenodiazol und fluorescierendem 2,1,3-Naphthoselenodiazol. Die direkte Cortisol-Bestimmung wird neben der Kondensation zu Cortisoldansylhydrazon beschrieben. Fluorescierende Testosteron-Derivate werden durch postchromatographische Derivatisierung mit (a) Isonicotinsäurehydrazid, (b) auf basischen Al2O3-Platten und (c) mit Ammoniumhydrogencarbonat sowie durch prächromatographische Derivatisierung mit Dansylhydrazin erhalten. Lecithin und Sphingomyelin werden mit MnCl2 und Carbamazepin durch Behandlung mit HCl-Dämpfen oder HClO4-Lösung zu Fluorescenz-Derivaten umgesetzt. Niedere flüchtige Carbonsäuren werden zu p-Brom-Phenacylestern, Naphthacylestern und DAPACAP-Estern umgesetzt.
