Summary
By a combination of multiple distribution methods with adsorption chromatography two new irritant and cocarcinogenic Euphorbia factors C and C′ have been isolated from latex of Euphorbia cooperi N. E. Br. By spectroscopic and chemical methods the factors were identified as tri- (C′) and di-esters (C), respectively, of the new diterpene alcohol 12-desoxy-16-hydroxyphorbol (II a). The diester C (II c) carries angelic acid (2-methyl-2-cis-butenic acid) at position 13 and isobutyric acid at position 16 of the parent alcohol II a. The triester C′ (II b) is the 20-acetate of the diester C (II c). Structure and position of the ester groups was confirmed by partial synthesis of the Euphorbia factors. Further two active compounds II f and II g were synthesized in an analogous manner.
The two new Euphorbia factors C and C′ show lower irritant activity than the croton oil factor A1 (TPA). The 13-[2-methyl-2-cis-butenoyl]-16-decanoyl-(II f) and -16-tetradecanoyl (II g) esters of 12-desoxy-16-hydroxy-phorbol, obtained by partial synthesis, however, exhibit irritant activities comparable to that of A1. As cocarcinogens the new Euphorbia factors are of much weaker activity than A1. Also, in a dose similar to that of A1 the diester II g obtained by partial synthesis is less cocarcinogenic than the croton oil factor.
Zusammenfassung
Mittels einer Kombination von Methoden der multiplikativen Verteilung und der Adsorptionschromatographie konnten zwei neue hautreizende und cocarcinogene Euphorbiafaktoren C und C′ aus Latex von Euphorbia cooperi N. E. Br. isoliert und rein dargestellt werden. Durch spektroskopische und ehemische Methoden wurden die Faktoren als Tri- (C′) bzw. Di-ester (C) des neuen Diterpenalalkohols 12-Desoxy-16-hydroxyphorbol (II a) charakterisiert. Der Diester C (II c) enthält einen Angelicasäurerest (2-Methyl-2-cis-butensäurerest) in Position 13 und einen Isobuttersäurerest in Position 16 des Grundalkohols II a. Der Triester C′ (II b) ist das 20-Acetat des Diesters C (II c). Die Struktur und Position der Estergruppen wurde durch Partialsynthese der Euphorbiafaktoren bestätigt. Zwei weitere Wirkstoffe wurden auf analogem Wege dargestellt.
Die hautreizende Wirkung der neuen Euphorbiafaktoren C und C′ ist geringer als die des Crotonölfaktors a1 (TPA). Die partialsynthetisch gewonnenen 13-[2-Methyl-2-cis-butenoyl]-16-dekan-(II f) bzw. -16-tetradekansäure-ester (II g) des 12-Desoxy-16-hydroxy-phorbols sind dagegen in ihrer hautreizenden Wirkung mit dem Crotonölfaktor A1 vergleichbar. Verglichen mit A1 zeigen die neuen Euphorbiafaktoren eine sehr geringe cocarcinogene Wirkung. Auch der stark hautreizende, partialsynthetische Di-ester II g ist bei etwa gleicher Dosierung wie A1 cocarcinogen weniger wirksam als der Crotonölfaktor.
