Abstract

Protein and peptide samples that require sequencing are, more often than not, received in biological buffers containing free amines, salts, and denaturants. Tris, guanidinium chloride, and urea are commonly used when purifying proteins, and these can interfere with the Edman degradation if present during sequencing. Additional sample manipulation, for example, dialysis, precipitation, or desalting by HPLC, is usually required to make the sample matrix compatible with the sequencing chemistries.
