Role of the IGF System in Breast Cancer Proliferation and Progression

Abstract

The insulin-like growth factor (IGF) family consists of two ligands (IGF-I and IGF-II), three receptors (IGFR1, IGFR2, and hybrid IGFR1/insulin receptor) and seven IGF binding proteins (IGFBP1-7). Similar to many other growth factor families such as epidermal growth factor (EGF) and platelet-derived growth factor (PDGF), the number and variety of effects that can be elicited by the IGF family are very diverse and continue to expand (1). In particular, the ability of different ligands, receptors, and binding proteins to crossreact and the ensuing variety of biologic effects make assigning specific roles for each member difficult. This is especially true for the IGFBPs; several years of research have still failed to elucidate the true function of these proteins in IGF action (2).
