Participation of Hepatic Parenchymal and Kupffer Cells in Chylomicron and Cholesterol Metabolism

Abstract

The administration of reticuloendothelial (RE) stimulants produced an increased growth and functional activity of RE cells. Parenchymal cell number, structure, and function, as indicated by BSP removal, and histological and electron-microscopic observations were unaltered in RE-stimulated rats. The RE hyperactive group manifested an increased intravascular removal of colloidal carbon which was associated with enhanced accumulation of carbon in Kupffer cells.

The technique of selectively and exclusively producing an RE hyper-functional state was employed to evaluate the metabolic behavior of labeled chyle, chylomicrons, and an artificial lipid emulsion, designated as “RE test lipid emulsion.” The intravascular removal of chyle and chylomicrons was not enhanced in RE-stimulated rats. The predominant uptake of injected chyle and chylomicrons was hepatic in nature with lung and spleen manifesting 1-3% removal. Cholesterol-containing chylomicrons demonstrated a behavior similar to that of triglyceride-containing chylomicrons and unlike colloidal carbon and the RE test lipid emulsion. Significant differences existed, however, between the organ uptake of cholesterol and triglyceride-labeled chylomicrons.

An artificial lipid emulsion was developed that possessed an intravascular behavior and an organ distribution which was comparable to colloidal and particulate materials which are removed by phagocytosis. This RE test lipid emulsion has been employed in the evaluation of RE function in experimental and clinical subjects.

Normal chylomicrons containing cholesterol or triglyceride are relatively immune to RE cell uptake and the hepatic mechanism of chylomicron removal involves essentially their removal by the parenchymal cell.
