Abstract

Historically, microorganisms have been identified using morphological and phenotypic criteria. As a consequence, classification frameworks have been constructed with limited and often fragmentary information. A major obstacle for microbiologists in the identification and classification of organisms is the ability to apply individual tests universally across all bacterial groups. For example, biochemical tests incorporating carbohydrates have been of great use in the affiliation of isolates to a particular taxon, but are of little value in the identification of anaerobic, assacharolytic organisms which use alternative biochemical pathways to derive their energy requirements (Busse et al., 1996; Vandamme et al., 1996). The outcome was that many taxa were created using a few poorly defined criteria and have been used as a repository for organisms that could not be classified with certainty.
