Abstract
The 5′-upstream regions of 5 different carp α-globin genes(No. 3 to No. 7) and four virus promoters(SV40, RSV, LTR, and SRα) were linked with the reporter gene, chloramphenicol acetyltransferase(CAT) gene. The transcriptional activity of carp 5′-upstream regions and virus promoter regions were examined by CAT assay after transfection into fish non-erythroid cell lines, which were established from the ovary of hybrid tilapia(TO-2), fin cells of colored carp(GF), and human erythroleukemia cell lines(K562 and HEL cell). The CAT assay shows that these 5′-upstream regions from carp α-globin genes possess the promoter function in these cell lines. In the GF cell line, the CAT activity was significantly stronger than in TO-2, K562 and HEL cell lines. The CAT activities in fish cell lines directed by the SRα promoter were higher than that of other virus promoters.
