Abstract

An analytical procedure based on isoelectric focusing was developed to identify contaminants in a nuclease preparation. Two categories of contaminants were visualized: protein impurities from the process background and modified forms of the enzyme. The potential of the analysis was tested on a fermentation with E. coli producing and secreting the enzyme, and using different purification procedures. Introduction of simple analyses to validate quality of a product at any stage in the developmental phase of a production can help to outline a production strategy.
