Zusammenfassung
Die molekulare Biologie der Karzinogenese und des Alterns zeigen bemerkenswerte Parallelen. Eine der ersten wegweisenden Beobachtungen war die replikative Seneszenz von primären humanen Zellen in Zellkultur. Demgegenüber zeigten zunächst nur Tumorzellen eine echte Immortalität in Kultur, jedoch auch genomische Instabilität. Die Telomer-Hypothese bot einen ersten Erklärungsansatz für die beobachteten Phänomene. Weitere Untersuchungen belegen jedoch, dass auch andere zelluläre Mechanismen, wie Autophagie und mitochondrialer Metabolismus, eine wesentliche Rolle beim Altern und der Karzinogenese spielen. Neuere Ergebnisse zeigen sogar eine aktive Rolle seneszenter Zellen in Entwicklung, Wundheilung sowie der Tumorentstehung und -progression.
