Abstract

The search for an endogenous inhibitor of Na+/K+-ATPase has been supported by the occurrence of digitalis-like cardiotonic steroids in plants and the presence of a highly conserved binding site for ouabain in mammalian tissues (5). The only precedent for cardiotonic steroids outside of the plant kingdom are the bufodienolides isolated from toads, although no glycosylated analogs were described (23). A substantial amount of indirect evidence supporting the existance of Na+/K+-ATPase inhibitory activity in human tissues has continued to fuel the search for a molecule of mammalian origin which would share at least some of the properties of the plant-derived glycosides, particularly as this compound(s) has been implicated in sodium and water homeostasis and the pathogensis of a prevalent human disease, hypertension (8).
