Abstract
The influx of ions into mitochondria may often be demonstrated most simply, in a qualitative manner, by observing the osmotic behavior of mitochondria in a solution containing the specific ion. Mitochondria (respiration-inhibited) are suspended in a salt solution which has the same osmolarity as the mitochondrial matrix compartment (about 300 mosmolar); any influx of salt will be accompanied by an influx of water and the mitochondria will consequently increase in volume. Mitochondrial swelling is accompanied by a decrease in the light scattering of the suspension and this may be monitored spectrophotometrically.
