Abstract

Karyotype instability is an inherent problem of in vitro cultures, since most cultures consist of a heterogeneous mass of cells in different states of differentiation and dedifferentiation. Haploid cultures, in spite of possible initiation from a uniform population of uni-nucleate haploid microspores or haploid protoplasts, exhibit similar types of instability. Because of their origin from single haploid cells they can provide a unique opportunity for studying the factors leading to variability. Cell cultures are also of importance for advances in somatic fusion and gene transfer into defined plant cells, since in vitro cultures and subsequent regeneration of plants are usually necessary. However, induced variability might interfere with directed and controlled genetic manipulation (Lorz and Brown 1986).
