Abstract

Poly (ADP-ribose) polymerase is a eukaryotic enzyme that uses NAD as substrate to synthesize homopolymers of ADP-ribose which are covalently linked to protein acceptors such as the enzyme itself (Carter and Berger, 1982). The enzyme is predominantly located in the nucleus in an inactive form; it is activated by DNA strand breaks that result from a variety of treatments that cause DNA damage (Berger, et al., 1979; Cohen and Berger, 1981). Treatment of cells with agents such as 3 aminobenzamide (3AB) to inhibit poly(ADP-ribose) polymerase interferes with the repair of DNA strand breaks and suggests that poly(ADP-ribose) synthesis is required for DNA repair (Durkacz, et al., 1980). Other studies suggest that poly(ADP-ribose) synthesis may also be involved in DNA replication and cell differentiation (Hayaishi and Ueda, 1977). Since many of these studies are based on the use of enzyme inhibitors which also affect other cellular process, the function of poly(ADP-ribose) polymerase has not yet been determined with certainty.
