Modulation of Estrogen Metabolism by Inducers of P-4501A2: Possible Application to the Chemoprevention of Breast Cancer

Summary

In these studies we have demonstrated that noninvolved mammary tissue obtained from a breast containing a tumor showed increased FTP binding activity of ras-P21and increased 16α-hydroxylation of estradiol relative to the level in normal breast tissue. In addition, a hyper response in the levels of both parameters was observed following treatment of the tumor-related breast tissue in organ culture with benzopyrene and/or fatty acids. The results suggest that the apparently normal breast tissue from a cancerous breast is biochemically different from breast tissue taken from a normal breast (mammoplasty). The shifts in these parameters after both treatments are in the direction associated with increased cancer risk.
