Diagnosis of Malignant Hyperthermia in Japan by the Skinned Fibre Test

Abstract

As a diagnostic test of MH susceptibility, the whole muscle caffeine halothane contracture test should be employed.

In order to obtain more definitive information about MH tests, calcium uptake into the SR, calcium release from the SR and calcium related functions of the contractile system should also be utilized using single chemically skinned fibres. Both the dose of halothane or of caffeine necessary to induce threshold contractures and calcium induced calcium release should be measured.

The skill necessary to manipulate a skinned fibre is one of the great barriers to adequate performance of the skinned fibre test. Well-trained and precise hands with patience are required. There are many advantages, however, which make this technical difficulty well worth surmounting.
