Abstract

Immunoglobulins (Igs) are the paradigm of a cancer marker because of their diagnostic, prognostic, and ontologic significance. The recognition that Igs serve uniquely as biomarkers of the neoplastic transformation of a cellular system that is committed to the synthesis and secretion of antibody began over 100 years ago when scientists discovered that an unusual urinary protein—the Bence-Jones protein—was associated with a malignant cellular disease of the bone marrow—multiple myeloma. The importance of Igs as cancer markers was extended by the discovery of other types of unusual proteins—the myeloma proteins and Waldenström macroglobulins—in the serum of patients with plasma-cell and certain related B-cell lymphocytic neoplasms.
