Abstract
“Epicytosine” or 5-methylcytosine was first noted as a component of calf thymus DNA in 1948 (Hotchkiss, 1948). The non-random distribution of 5-methylcytosine in DNA led to the suggestion that this minor base derived from post-replication modification of DNA (Romberg et al., 1959). Subsequent studies showed that DNA modification in prokaryotes is part of the host range restriction system (Arber, 1968) and is also involved in strand selection in mismatch repair of DNA (Radman and Wagner, 1984). Although DNA methylation may serve other functions in prokaryotes, attention has recently focused on DNA methylation in eukaryotic systems. Application of molecular biological techniques to analyses of eukaryotic DNA methylation has led to explosive growth of this field; almost 300 papers were published just in 1984. This paper will present an overview of only selected recent analyses of eukaryotic DNA methylation.
